New support from Grant Agency of Czech Republic

Grant Agency of Czech Republic will support our new project named Multi-FRET fluorescent probes responding to hydrolases.


The grant proposal was composed after recently developed quite new system for unambiguous resolution of one of two proteases or their mixture based on three fluorophores, (coumarine (DEAC), fluorescein (Fl), rhodamine B (RhB)) in the pH independent mode) with double FRET between them (see Figure).

The project is focused on synthesis of model systems for detection of three proteases or a protease and β-lactamase solely or in their mixture via fluorescence multi-FRET effect. For this purpose the synthetic protocol will be developed and the rules for evaluation of individual emissions to distinguish various combinations of proteases will be found and verified. The other part of the project is focused on development of the system for simultaneous detection of β-lactamase and a protease with a possibility to release a drug or an inhibitor. Within the project the concept of the new probes will be verified with various combinations of dyes and cleavable linkers. Possible simultaneous β-lactamase triggered releasing will be tested with various small compounds including a drug or a β-lactamase inhibitor.

The project is awarded for the years 2019-2021.
